Purification, characterization, DNA sequence and cloning of a pimeloyl-CoA synthetase from Pseudomonas mendocina 35.
A pimeloyl-CoA synthetase from Pseudomonas mendocina 35 was purified and characterized, the DNA sequence determined, and the gene cloned into Escherichia coli to yield an active enzyme. The purified enzyme had a pH optimum of approximately 8.0, Km values of 0.49 mM for pimelic acid, 0.18 mM for CoA and 0.72 mM for ATP, a subunit Mr of approximately 80000 as determined by SDS/PAGE, and was found...
متن کاملCloning, Expression, and Purification of Glutamine Synthetase from Clostridium acetobutylicum.
A glutamine synthetase (GS) gene, glnA, from the gram-positive obligate anaerobe Clostridium acetobutylicum was cloned on recombinant plasmid pHZ200 and enabled Escherichia coli glnA deletion mutants to utilize (NH(4))(2)SO(4) as a sole source of nitrogen. The cloned C. acetobutylicum gene was expressed from a regulatory region contained within the cloned DNA fragment. glnA expression was subje...
متن کاملProduction of Non-Ribosomal Peptide Synthetase (NRPS)- Dependent Siderophore by Aeromonas Isolates
Background: Aeromonas species are Gram-negative ubiquitous bacteria, facultative anaerobic rods that infect both invertebrates and vertebrates. Various fish species develop hemorrhagic disease and furunculosis due to Aeromonas spp. Aeromonas strains generate certain active compounds such as siderophores, which are the final products of non-ribosomal peptide synthetase (NRPS) activity. The prese...
متن کاملDetection of nonribosomal peptide synthetase genes in Xylaria sp. BCC1067 and cloning of XyNRPSA.
Nonribosomal peptides, synthesized by nonribosomal peptide synthetases (NRPS), are an important group of diverse bioactive fungal metabolites. Xylaria sp. BCC1067, which is known to produce a variety of biologically active metabolites, was studied for gene encoding NRPS by two different PCR-based methods and seven different NRPS fragments were obtained. In addition, screening a genomic library ...
متن کاملTasiamide, a cytotoxic peptide from the marine cyanobacterium Symploca sp.
The acyclic peptide tasiamide (1) has been isolated from the marine cyanobacterium Symploca sp. The planar structure and absolute stereochemistry of the peptide were deduced by 2D NMR techniques and chiral HPLC. Tasiamide (1) was cytotoxic against KB and LoVo cells with IC(50) values of 0.48 and 3.47 microg/mL, respectively.
